There isn't really a set phrase for that. It will change from instance to instance.

If you list a few specific examples of what you're trying to say, I can translate them for you.

Like for instance.... Can I be Your Ninja? Kunaichi is the word for a female ninja i belive.

the way you say 'may I' is -te form + mo ii desu ka. 'May I be' could be 'date mo ii da?', although in your case you would probably use the verb 'naru' - to become, which would get you 'anata no kunoichi ni natte mo ii desu ka'.
